Clinical Information
HLA Class I genotyping defines the antigens of the HLA-A, B and C loci of the Major Histocompatibility Complex found on the short arm of chromosome 6. Using DNA extracted from leukocytes, HLA-A, B, and C alleles are amplified via PCR using sequence-specific primers. After amplification, the specific HLA antigens are determined by gel electrophoresis. HLA genotyping serves an important role in transplantation and disease association studies.
Specimen Requirements
Specimen Collection Criteria
- Collect: Two Lavender-top EDTA tubes OR one Pink-top tubes OR one Yellow-top ACD tubes.
- Invert several times to mix well.
Physician Office/Draw Specimen Preparation
- Do not refrigerate or freeze specimen. Maintain whole blood at room temperature (20-26°C or 68-78.8°F) prior to transport. Transport to the Laboratory within 72 hours of collection.
Preparation for Courier Transport
- Transport: Whole blood at room temperature (20-25°C or 68-77°F).
Rejection Criteria
- Unlabeled specimens.
- Specimens drawn in clot, SST, or Sodium Heparin tubes.
In-Lab Processing
- Maintain specimen at room temperature (20-26°C or 68-78.8°F) until testing.
